Acetyl-6-formylpterin (Ac-6-FP) is a hapten that stabilizes MR1 on antigen-presenting cells, forming an MR1:A6FP complex that fails to activate MAIT cell TCR, thereby acting as a competitive inhibitor of MAIT cell activation. Acetyl-6-formylpterin exacerbates house dust mite-induced airway hyperresponsiveness and elevates circulating IgE levels. Acetyl-6-formylpterin reduces systemic MAIT cell activity and decreases the frequency of IL-17A+ and IFNγ+ MAIT cells in the spleen. Acetyl-6-formylpterin is useful for the study of allergic asthma.

Did not alter HDM-induced pulmonary inflammation, granulocyte or lymphocyte counts, cytokine production by non-T cells or conventional T cells, or mucus production.
Further increased HDM-induced serum IgE levels without affecting IgA, IgM, or IgG1 levels.
Did not aggravate HDM-induced AHR in IgE-deficient mice.
Failed to increase AHR in B cell-deficient μMT mice.

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (10.72 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.

Ovalbumin-Induced Allergic Airway Inflammation
Ovalbumin-induced allergic airway inflammation is a mouse model in which systemic sensitization to ovalbumin, usually with aluminum hydroxide adjuvant, is followed by airway ovalbumin challenge to induce allergic airway inflammation, eosinophil recruitment, mucus production, serum antigen-specific IgE, Th2 cytokine responses, and airway hyperresponsiveness to methacholine. The model is used to study allergen-driven airway inflammation and asthma-like immune responses, but it does not reproduce every feature of human asthma. The main readouts are bronchoalveolar lavage fluid cellularity, lung histopathology, airway hyperresponsiveness, serum OVA-specific IgE, and cytokines such as IL-4, IL-5, and IL-13 in bronchoalveolar lavage fluid or lung samples. Eosinophilia and Th2 cytokines reflect allergic type 2 inflammation, while methacholine responsiveness provides a functional airway-reactivity endpoint.
