dATPαS

dATPαS (2'-Deoxyadenosine 5'-O-(1-thiotriphosphate)) is incorporated by Bst more slowly than the canonical 2'-Deoxyadenosine-5'-triphosphate (dATP) (HY-136648), with a relative rate lower than that of TTPαS and dCTPαS (HY-W784574) and higher than that of dGTPαS (HY-W784575)^[1].
dATPαS (1-50 µM; 0.02-4 s) is incorporated in both Sp and Rp forms by wild-type Pol β, with a preference for Sp-dATPαS, and the Sp/Rp specificity ratios in the presence of Mg²⁺, Mn²⁺, and Cd²⁺ are 57.5, 7.6, and 21, respectively^[2].
The stereoselectivity of dATPαS (5 mM Mg²⁺; 1 mM Mn²⁺; 0.1 mM Cd²⁺) is relaxed by the D276R Pol β mutant, with Sp/Rp ratios of 1.5, 3.7, and 1.5 in the presence of Mg²⁺, Mn²⁺, and Cd²⁺, respectively, and its reactivity toward Rp-dATPαS is increased 5-fold relative to wild-type Pol β^[2].
dATPαS (1 mM; 24-26 h) binds to the active site in human pol β-DNA 7bp crystals containing 5 mM Mn²⁺, but does not support nucleotide transfer, and preferentially adopts the R α-thiophosphate configuration^[3].

dATPαS serves as a dNTP substrate in the enzymatic photoluminescence cascade used for cell-free pyrosequencing of bisulfite-converted genomic DNA to quantify CpG methylation^[4].
