Matuzumab (EMD 72000) is a humanized anti-EGFR monoclonal antibody that blocks EGFR activation and downstream signaling, inhibits tumor growth. Deglycosylation and the possible elimination of ADCC activity can reduce the in vivo anti-tumor activity of Mattuzumab.

Matuzumab (10 ng, 4-72 h) can exert a powerful anti-tumor ADCC effect in the target monolayer cells of head and neck squamous cell carcinoma (SCCHN)[2].
Matuzumab was enzymatically de-glycosylated to eliminate its ADCC activity, but the de-glycosylated antibody maintained its ability to bind to A431 tumor cells, block EGFR signaling, and inhibit growth in vitro[1].
