DYRK2 ligand 2 (compound 8A2) binds to purified DYRK2 protein with a K_d value of 4.7 μM, and inhibits the kinase activity of purified DYRK2 with an IC₅₀ of 19 nM^[1].
DYRK2 ligand 2 can be used to synthesize PROTAC DYRK2 degrader-2 (HY-186287). PROTAC DYRK2 degrader-2 is a PROTAC-based DYRK2 degrader derived from curcumin analog A3. It recruits the CRBN E3 ligase to mediate the ubiquitination of DYRK2, leading to degradation via the ubiquitin-proteasome system^[1].
