TRX-518 is a humanized agylcosyl IgG1 anti-GITR mAb, , and is a GITR agonist. TRX-518 binds to the extracellular domain of human GITR, abrogates Treg-mediated suppression. TRX-518 increases effector T cell activation and pro-inflammatory cytokine production, reduces circulating and intratumor Treg frequencies. TRX-518 destabilizes Treg phenotype via Foxp3 downregulation and T-bet upregulation. TRX-518 can be used for the research of solid tumors[1][2][3].

TRX-518 destabilizes the phenotype of Tregs derived from healthy human donor CD4+ T cells by down-regulating Foxp3 and enhancing Th1-lineage transcription factor expression[2].

Flow cytometric analysis of 1X106 Jurkat cells with TRX-518 (HY-P991180, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -

Naïve CD4+ T-cell subset differentiation/polarization is an in vitro assay in which purified naïve CD4+ T cells are activated through TCR and CD28 costimulation and cultured with defined cytokines and neutralizing antibodies to generate Th0, Th1, Th2, Th17, or induced Treg-like populations. Differentiation is detected by subset-associated cytokines and transcription factors: IFN-γ/T-bet for Th1, IL-4/GATA3 for Th2, IL-17A/RORγt for Th17, and Foxp3 for induced Treg cells. The assay readout is usually generated by intracellular cytokine staining after restimulation, transcription-factor staining by flow cytometry, ELISA of secreted cytokines, or gene-expression analysis. The result reflects cytokine-directed lineage commitment or polarization rather than antigen-specific immune protection by itself.
