GTP-γ-S
GTPγS (Guanosine 5'-[γ-thio]triphosphate) is a G-protein activator that protects proteins from proteolytic degradation, stimulates GLUT4 translocation in a tyrosine kinase-dependent manner, stimulate phospholipases and induce actin polymerization. GTPγS to couple with G- protein α, to study its effect on kinase activity. GTPγS acts as a component of lysis buffer.

Biological Activity
GTPγS (30-100 μM; 1-3 h) maintains Rab27a in a GTP-bound state that supports its indirect association with melanocyte myosin Va (MC MV) and MC MVΔD in B16 F10 melanocyte lysates, whereas GDP reduces this association[1].
GTPγS (1 mM; 20 min) stimulates glucose transport in electrically permeabilized rat primary adipocytes via a microtubule-independent mechanism[2].
GTPγS (1 mM; 20 min) induces GLUT4 translocation to the plasma membrane in electrically permeabilized rat primary adipocytes with more residual cytoplasmic GLUT4 signals than insulin[2].
GTPγS (1 mM; 20 min) does not disrupt the association of tubulin with GLUT4-containing membranes in electrically permeabilized rat primary adipocytes, in contrast to insulin[2].
GTPγS (10 μM) defines nonspecific [³⁵S]GTPγS binding in coronal sections of rat brain (including dorsal raphe nucleus, median raphe nucleus, and forebrain areas)[3].
GTPγS (10 μM; 15 min) formed Gαi2GTPγS that enhances recombinant human Src tyrosine kinase activity with an ED50 of 20 nM, while formed GαzGTPγS reduces this stimulatory effect[4].
