CAN10
Based on 1 Customer Validation
CAN10 is is a humanized monoclonal antibody inhibitor targeting interleukin 1RAP (IL1RAP). CAN10 blocks IL-1, IL-33, and IL-36 signalling by targeting IL1RAP, preventing downstream proinflammatory and profibrotic responses. CAN10 reduces dermal and pulmonary fibrosis. CAN10 can be used for the research of immune system diseases, such as systemic sclerosis.

CAN10 effectively impairs the proinflammatory and profibrotic effects of IL1RAP-related cytokines (IL-1, IL-33, and IL-36) in human umbilical cord endothelial cells and human dermal fibroblasts, which are critical to systemic sclerosis pathophysiology[1].

Result:Ameliorated clinical signs of cutaneous cGvHD and reduced mean composite scores.
Ameliorated cGvHD-induced weight loss more effectively than the positive control reference drug.
Reduced dermal thickening, accumulation of hydroxyproline, and myofibroblast differentiation compared with vehicle- or isotype control-treated mice.
Reduced Ashcroft scores and hydroxyproline content in lung tissue compared with vehicle- or isotype control-treated mice, with numerical decreases in collagen-covered lung area.

Flow cytometric analysis of 1X106 HepG2 cells labeling IL1R3 with CAN10 (HY-P991267, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 for an hour at 4℃. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
