MU876
Based on 1 Customer Validation
MU876 is a MUS81 inhibitor (IC50 0.52 μM). MU876 inhibits Endonuclease activity and disrupts its binding to branched DNA substrates. MU876 inhibits homologous recombination and break-induced replication DNA repair without affecting non-homologous end joining. MU876 induces the accumulation of micronuclei, telomere-free chromosome ends, and DNA damage foci. MU876 sensitizes cancer cells to the cytotoxicity of Cisplatin (HY-17394). MU876 does not induce apoptosis after short-term exposure, nor does it significantly perturb cell cycle distribution at low concentrations.

In Vitro
MU876 inhibits BIR in U2OS BIR-GFP cells with an IC50 of 0.24 μM, and inhibits HR with an IC50 of 0.5-1 μM, without impairing NHEJ[1].
MU876 (0.25-0.5 μM; twice weekly for nearly 3 weeks) inhibits the proliferation of CAL51 WT cells over nearly three weeks[1].
MU876 (3-12 μM; 48 h) increases micronucleus formation in U2OS cells[1].
MU876 (72 hours) increases γH2AX foci in U2OS cells after 72 hours[1].
MU876 (maximum concentration 1 μM, 2-fold serial dilution) reduces clonogenic survival of U2OS cells[1].
MU876 enhances Cisplatin (HY-17394)-induced loss of viability in U2OS, HEK293, and CAL51 WT cells, but has no such effect on CAL51 MUS81−/− cells[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
