Alnuctamab (72 h), when co-cultured with T cells and BCMA+ myeloma cell lines (H929, OPM-2, L363), in combination with CELMoD drugs (Pomalidomide (HY-10984), Mezigdomide (HY-129395) and Iberdomide (HY-101291)), enhances T cell-mediated tumor cell killing efficacy, as demonstrated by reduced IC50 values and increased clearance rate of multiple myeloma cells[1].

In Vivo

Alnuctamab, when combined with Mezigdomide (HY-129395) in immunodeficient mice implanted with H929 myeloma cells, significantly inhibits tumor growth, enhances the infiltration of T cells (especially CD8+ cells) into tumors, and increases the expression of T cell activation markers (such as CD69) within tumors[1].

Biological Activity
  • Immobilized CD3 epsilon/CD3e Protein can bind Alnuctamab. The EC50 for this effect is 0.728 ng/mL.
  • Immobilized BCMA/TNFRSF17 Protein, Human can bind Alnuctamab. The EC50 for this effect is 0.051 ng/mL.
  • Flow Cytometry analysis of K562 cells labelling BCMA (red) with Alnuctamab (HY-P99430). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then cells were stained with the primary antibody at 1200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 11,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
