Cell division cycle 6 (CDC6) is essential for the initiation of DNA replication in eukaryotic cells and is closely related to cell-cycle regulation in cancer[1]. Mechanistically, CDC6 acts through cell-cycle events and the PI3K-AKT pathway, linking DNA replication programs with proliferation, migration, invasion, apoptosis resistance, and immune infiltration[1][3]. In disease models, CDC6 expression was significantly upregulated in PCOS and endometrial cancer, and CDC6 knockdown reduced endometrial cancer cell proliferation, migration, and invasion while increasing apoptosis[1]. In small cell lung cancer datasets, CDC6 appeared among hub genes enriched in cell cycle, DNA replication, oocyte meiosis, and mismatch repair pathways[2]. Compared with related cell-division-cycle markers such as CDC20, CDC6 is supported here by direct functional knockdown evidence in endometrial cancer and hepatocellular carcinoma models[1][2][3]. For experimental applications, CDC6 currently has stronger evidence as a diagnostic, prognostic, immunotherapy, and targeted-therapy biomarker than as a validated agonist or inhibitor target[3].
