NC1 is a selective non-competitive and allosteric lymphoid-specific tyrosine phosphatase (LYP) inhibitor, with a Ki value 4.3 μM. NC1 allosterically regulates LYP/PTPN22 activity by restricting WPD loop movement. NC1 inhibits LYP activity in lymphoid T cells and enhances T-cell receptor signaling. NC1 can be used for the research of autoimmune diseases.

Biological Activity
NC1 (0-10 μM) non-competitively inhibits wild-type LYP with a Ki of 4.3 ± 0.3 μM in a pNPP hydrolysis biochemical assay[1].
NC1 shows reduced binding affinity for LYP mutants with alterations to residues T36, R266, H196, F28, D197, and C129, indicating these residues contribute to NC1's interaction with LYP[1].
NC1 (20 μM; 45 min pre-incubation) effectively and specifically enhances TCR signaling in Jurkat T cells by inhibiting endogenous LYP activity, as demonstrated by increased LCK and ERK phosphorylation that is abolished by LYP knockdown[1].
