Description

Dianellin is an orally active anti-malarial agent found in Kniphofia foliosa. Dianellin binds to Plasmodium falciparum plasmepsin II and shows anti-malarial activity in mice. Dianellin can be used for the research of malaria[1].

In Vitro

Dianellin weakly binds to Plasmodium falciparum plasmepsin II and does not bind to Plasmodium falciparum pfLDH[1].

In Vivo

Dianellin (25-100 mg/kg; i.g.; daily; 4 days) exhibits dose-dependent in vivo anti-malarial activity against Plasmodium berghei in Swiss albino mice, with an ED50 of 92.31 mg/kg and up to 53.77% parasitaemia suppression at the highest tested dose[1].

Animal Model: Swiss albino (5-6 weeks old, 20-25 g, male or female, intraperitoneally inoculated with Plasmodium berghei ANKA strain-infected erythrocytes)[1]
Dosage: 25, 50, 100 mg/kg
Administration: p.o.; daily; 4 days
Result: Achieved 29.83% parasitaemia suppression and a mean survival time of 7.6 days at 25 mg/kg/day.
Achieved 44.04% parasitaemia suppression and a mean survival time of 8.2 days at 50 mg/kg/day.
Achieved 53.77% parasitaemia suppression and a mean survival time of 8.2 days at 100 mg/kg/day.
Demonstrated statistically significant suppression compared to negative control (p < 0.001).
Had a median effective dose (ED50) of 92.31 mg/kg.
