dTAG-TRIMTAC
dTAG-TRIMTAC is a PROTAC-like degrader targeting TRIM21. dTAG-TRIMTAC forms a ternary complex with TRIM21 and target proteins to drive degradation of oligomeric/assembled substrates via TRIM21 clustering-based activation, with degradation dependent on endogenous TRIM21. dTAG-TRIMTAC can be used for the research of neurodegenerative disease and inflammatory disease.

dTAG-TRIMTAC (5 μM) selectively degrades oligomeric Myddosome-associated Myd88-GyrB-mEGFP-FKBP(F36V) in Coumermycin-stimulated U2OS cells, with no activity against monomeric Myd88-GyrB-mEGFP-FKBP(F36V) in unstimulated cells[1].
dTAG-TRIMTAC (0.16-2.5 μM; 8-48 h) selectively degrades oligomeric Necrosome-associated FKBP(F36V)-mEGFP-RIPK3 in T/S/Z-stimulated RPE-1 cells, with no activity against monomeric FKBP(F36V)-mEGFP-RIPK3 in unstimulated cells, and partially inhibits necroptotic cell death[1].
dTAG-TRIMTAC (0.11-10 μM; 1 h pre-incubation) dose-dependently inhibits seeded P301S tau aggregation in HEK293 reporter cells, achieving up to 60% inhibition[1].
