Eluvixtamab (AMG-330) is a bispecific T-cell engager (BiTE) antibody targeting CD33 and CD3, with a Kd of 5.1 nM and 8 nM for human and cynomolgus monkey CD3, respectively, and a Kd of 8 nM and 8.6 nM for human and cynomolgus monkey CD33, respectively. Eluvixtamab links T cells to CD33-expressing acute myeloid leukemia (AML) cells, triggers T-cell activation, and induces inflammatory adaptive immune escape characterized by PD-L1 upregulation in AML cells. Eluvixtamab is applicable to research related to acute myeloid leukemia.

Eluvixtamab (AMG 330) (5 ng/mL; 3-36 days) mediates T-cell-dependent lysis of primary AML cells ex vivo, with lysis kinetics dependent on initial E:T ratio, CD33 expression level, and the proportion of memory T cells in the effector population; prolonged incubation compensates for low initial E:T ratios or low CD33 expression[2].
Eluvixtamab (AMG 330) (5 ng/mL; 1-8 days) induces significant upregulation of PD-L1 on primary AML cells and PD-1 on autologous T cells ex vivo, with PD-L1 upregulation more pronounced on AML cells than T cells[2].
Eluvixtamab (AMG 330) (5 ng/mL; 2-18 days) has its mediated lysis of primary AML cells, T-cell proliferation, and IFN-γ secretion ex vivo significantly augmented by blockade of the PD-1/PD-L1 axis, with the greatest enhancement observed at low effector-to-target ratios[2].

Eluvixtamab (ex vivo pre-culture) pre-treatment of PDX AML cells completely prevents leukemia initiation in NSG mice, resulting in 100% survival over the observation period[2].

Flow cytometric analysis of 1×106 Jurkat cells labelling CD3E (red) with Eluvixtamab (HY-P99910). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -
