Histaprodifen
Histaprodifen is a selective histamine H1 receptor agonist that preferentially recruits β-arrestin2 over β-arrestin1. Histaprodifen activates phosphoinositide signaling and partially stimulates Gq-catalyzed GTP hydrolysis, stimulates astrocyte NGF secretion in a concentration-dependent manner, and produces guinea pig ileum/aorta contraction, rat aorta endothelium-dependent relaxation, and vascular hypotensive effects. Histaprodifen can be used for research on neurological diseases and cardiovascular diseases.

In Vitro
Histaprodifen stimulates NGF secretion in primary cultures of neonatal rat cortical astrocytes with a pD2 of 8.49 and an Emax of 84.6% relative to histamine[2].
Histaprodifen (compound 2) binds to recombinant hH1R, bH1R, rH1R, and gpH1R in Sf9 membranes with pKi values of 6.47, 6.09, 6.66, and 6.38, respectively[3].
Histaprodifen (compound 8a) (3-30 µM; 0.5-10 min) is a potent agonist of guinea pig ileum H1 receptors, with a pEC50 of 6.74[5].
Histaprodifen is a weak H3 receptor antagonist in guinea pig ileum, with a pA2 of 5.51[5].
Histaprodifen exhibits weak M3 receptor antagonist activity in guinea pig ileum, with a pA2 of 5.55[5].
Histaprodifen (compound 5) is a full H1 receptor agonist in isolated guinea pig ileum, with a pEC50 of 6.7[6].
