KVS0001 is a selective SMG1 inhibitor. KVS0001 elevates the expression of transcripts and proteins resulting from truncating mutations. KVS0001 increased the presentation of immune-targetable HLA class I-associated peptides from nonsense-mediated decay (NMD)-downregulated proteins on the surface of cancer cells. KVS0001 exerts anti-tumor properties and can be studied in research for NMD-related diseases, including cancer and inherited diseases.

Biological Activity
KVS0001 (10 nM-1 μM) inhibits SMG1 protein more than any other 246 protein or lipid kinases[1].
KVS0001 (0.2-5 μM) is bioactive in nanomolar range and subverts the NMD-mediated downregulation of truncating mutant transcripts and proteins in NCI-H358 and LS180 cells[1].
KVS0001 causes significant increase in IFN-γ release in NCI-H716_A24 and NCI-H2228 cells in the presence of KVS-BI043 bispecific antibody and T cells, as well as leading to significant killing of the target cancer cells[1].
KVS0001 (5 μM) significantly increases RNA in six of the twelve truncating mutation-containing genes and five of the six expression controlled by NMD genes in murine RENCA (renal cancer) and LLC (lung cancer)[1].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (4.32 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
