In Vitro

3-Hydroxybenzoic acid (0.01-10⁵ μM) acts as an agonist for Hydroxycarboxylic acid receptor 1 (HCA1) (GPR81) and HCA2 (GPR109A)^[1].
3-Hydroxybenzoic acid (0.1-10⁵ μM) inhibits cAMP accumulation in SK-N-MC cells which stably expressing human HCA1^[1].
3-Hydroxybenzoic acid (100-1000 μM, 2 h) inhibits free fatty acid and glycerol release in mature adipocytes isolated from mouse subcutaneous and epididymal fat^[1].

In Vivo

3-Hydroxybenzoic acid protects mice (3-300 mg/kg; p.o.; 7 times in 12 days) and rats (20 mg/kg; p.o.; 7 times in 12 days) against stress triggered alterations in body weights and core temperatures, acts as stress response desensitizers^[2].
3-Hydroxybenzoic acid (20 mg/kg; p.o; 9 times in 12 days) possesses anti-inflammatory and analgesic activities in wistar rats^[3].

Animal Model:

Adult male Wistar rats (150 g)^[2]
Dosage:

20 mg/kg
Administration:

Oral gavage (p.o.); 7 times in 12 days
Result:

Could counteract mild unpredictable stress triggered body weight losses and slight elevation of basal rectal temperatures in rats.
Decreased stress induced hyperthermia.
Significantly lower mean plasma glucose and cortisol and higher insulin levels were observed.

Male Swiss mice (20 g) with occasional foot shock stress^[2]

3, 10, 30, 100, 300 mg/kg
Administration:

Oral gavage (p.o.); 7 times in 12 days
Result:

Remained constant up to the 10th day of the experiment, but after 30 mg/kg or more daily doses mean body weights of the treated groups increased dose dependently on the last observational days.
Intermittent foot-shock induced elevations of core temperatures were antagonized significantly.
Decreased stress induced hyperthermia.
Had statistically significant effects on immobility period in tail suspension test on male mice.
Statistically significant prolongation of pentobarbitone induced sleep duration was observed after 30 mg/kg and higher daily doses.

Stock Solution Working Solution

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 2.5 mg/mL (18.10 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Protocol 2

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

Solubility: 2.5 mg/mL (18.10 mM); Suspended solution; Need ultrasonic

This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Protocol 3

Add each solvent one by one: 10% DMSO 90% Corn Oil

Solubility: ≥ 2.5 mg/mL (18.10 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
