Isonicotinamide
Based on 1 Customer Validation
Isonicotinamide is a structural isomer of Nicotinamide (HY-B0150). Isonicotinamide promotes the activity of the NAD+-dependent deacetylase Sir2 in yeast and affects NAD+ homeostasis and nucleotide salvage synthesis pathways. Isonicotinamide can also serve as an intermediate for the synthesis of active drugs.

In Vitro
Isonicotinamide (25-125 mM; day 2) high concentrations above 50 mM inhibit BY4741 growth, with 125 mM causing strong early growth reduction[1].
Isonicotinamide (25-125 mM; 2 days) sensitivity across the yeast knockout collection is dose-dependent, with 150-326 mutants sensitive at 25-125 mM and 341 genes from the 25-75 mM conditions used for pathway enrichment[1].
Isonicotinamide (25-75 mM; day 2) sensitivity occurs in htz1Δ and SWR1 complex mutants, with htz1Δ sensitive at 25 mM[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
In Vivo
Isonicotinamide (976 mg/kg/day; intraperitoneal; daily; 10 days) does not prolong survival in L1210G leukemia as a single agent, but in combination with ATDA at 33.7 or 16.85 mg/kg/day it increases survival time[2].

With ATDA 7.5 mg/kg/day at 30.5 mg/kg/day, tumor weight was 12 mg and inhibition was 98%.
With ATDA 7.5 mg/kg/day at 15.25 mg/kg/day, tumor weight was 266 mg and inhibition was 63%.
With ATDA 7.5 mg/kg/day at 60 mg/kg/day, tumor weight was 27 mg and inhibition was 96%.
With ATDA 3.75 mg/kg/day and 60 mg/kg/day, tumor weight was 256 mg and inhibition was 58%.
With ATDA 1.875 mg/kg/day and 60 mg/kg/day, tumor weight was 325 mg and inhibition was 47%.
In the separate B82T experiment, ATDA 7.5 mg/kg/day with 122 mg/kg/day produced tumor weight 2 mg and 100% inhibition.
In the separate B82T experiment, ATDA 7.5 mg/kg/day with 61 mg/kg/day produced tumor weight 34 mg and inhibition 96%.

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (40.94 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 5 mg/mL (40.94 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.

Working solution concentration: 0.22 mg/mL
Method for preparing stock solution: mg drug dissolved in μL DMSO. Stock solution concentration: mg/mL.
1. Take μL DMSO stock solution;
2. Add μL .
μL , mix evenly;
3. Then add μL Tween 80, mix evenly;
4. Then add μL
