KM91104 is a cell-permeable V-ATPase a3-b2 inhibitor (IC50 = 2.3 µM). KM91104 reduces the metabolic activity, cell proliferation capacity and V-ATPase subunit protein expression levels of primary human hepatic stellate cells, increases intracellular ATP levels and decreases cytoplasmic pH. KM91104 reduces TLR4 expression on the surface of oligodendrocyte precursor cells, blocks the ENV-TLR4 interaction, and reverses oligodendrocyte myelination defects induced by ENV protein.

Biological Activity
KM91104 (1-10 nM; 48 h) reduces metabolic activity, cell proliferation capacity and v-ATPase subunit protein expression levels in primary human hepatic stellate cells, increases intracellular ATP levels and decreases cytoplasmic pH[1].
KM91104 (0.5 μM; 1-6 d) reverses the differentiation inhibition of primary rat cortical oligodendrocyte precursor cells (OPCs) mediated by pHERV-W ENV, restores the expression of myelin markers (MBP, MOG) and the mature oligodendrocyte marker CC1 to baseline levels, and does not affect cell viability[2].
KM91104 (0.5 μM; 1 d) reduces the expression level of TLR4 on the surface of primary rat cortical oligodendrocyte precursor cells (OPCs), blocks the direct binding of TLR4 to envelope (ENV), and thereby inhibits the ENV-mediated TLR4 signaling cascade[2].
KM91104 (0.5 μM; 13 d) reverses the inhibitory effect of pHERV-W ENV on myelination in embryonic rat cortical neuron/oligodendrocyte co-culture systems, and restores the proportion of myelinating oligodendrocytes to baseline levels[2].

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (9.18 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
