TMB-8 is a benzoate ester compound. TMB-8 blocks intracellular calcium (Ca) release, catecholamine secretion, and cholinergic/CCK-stimulated amylase release, while stimulating pancreatic hormone release and, under hyperthermia conditions, increasing cytosolic calcium and hyperthermic cell killing. TMB-8 is used in research on chemotherapy-induced painful peripheral neuropathy, human promyelocytic leukemia, and mouse breast cancer.

Anti-cancer

TMB-8 (10-100 μM; 4 days) inhibits HL-60 cell proliferation only at 100 μM while maintaining viability above 85%^[8].
TMB-8 (10-100 μM) alone induces NBT-positive differentiation of HL-60 cells^[8].

Carbohydrate Metabolism

TMB-8 dose-dependently stimulates the release of SRIF, IRG, and IRI from isolated rat pancreatic islets under 2.5 mM glucose conditions, and stimulates SRIF and IRG release without altering IRI under 10 mM glucose conditions^[5].

Cardiovascular Regulation

Renal Regulation

TMB-8 inhibits angiotensin II- and K⁺-stimulated aldosterone secretion in dispersed bovine adrenal zona glomerulosa cells, with ID₅₀ values of 46 µM and 28 µM, respectively^[10].

Others

TMB-8 concentration-dependently inhibits catecholamine secretion from isolated perfused rat adrenal glands evoked by Ach, excessive K⁺, DMPP, McN-A-343 (HY-107648), BAY-K 8644 (HY-10588), and 1,3,7-Trimethylxanthine, and almost completely blocks it at 100 μM^[3].
TMB-8 inhibits 1 μM Carbachol (Carbamoylcholine chloride) (HY-B1208)-stimulated amylase release, producing 57% inhibition at 10 μM, without affecting basal or 0.1 μM secretin-stimulated release^[4].
TMB-8 non-selectively inhibits parasympathetic neuromuscular transmission in guinea pig ileum and contractions induced by ACh, Nicotine, DMPP, KCl, and BaCl₂, with ID₅₀ values of 3.8 × 10^-6 to 14.4 × 10^-6 M, but BaCl₂ antagonism is competitive^[7].
TMB-8 directly inhibits protein kinase C activity supported by Ca²⁺ and phosphatidylserine in the cytosol of partially purified bovine adrenal zona glomerulosa cells in a dose-dependent manner^[10].

Cell Proliferation Assay^[8]
