trans-Ned 19 is a NAADP antagonist and TPC blocker. trans-Ned 19 suppresses the calcium signal and the rat aorta relaxation in response to low histamine concentrations. trans-Ned 19 increases the spontaneous acrosome reaction rate, alleviates anti-CD3 mAb-induced intestinal inflammation, and improves kidney damage in mice with nephrotoxic serum nephritis.

Biological Activity
NAADP, TPC, Calcium Channel[1]
trans-Ned 19 (25-100 μM; 1 h) inhibits T cell receptor (TCR)-stimulated Ca2+ signaling, cell activation and proliferation in primary naive CD4+ T cells from C57BL/6N mice[1].
trans-Ned 19 (25-50 µM; 10 min) inhibits Norepinephrine (HY-13715)-induced Ca2+ elevation in rat aortic smooth muscle cells in a concentration-dependent manner[2].
trans-Ned 19 (10 µM) causes lysosomal cholesterol accumulation in human umbilical vein endothelial cells, inhibits CD63 transport from late endosomes/lysosomes to Weibel-Palade bodies, and reduces the amount of CD63 associated with Weibel-Palade bodies[3].
trans-Ned 19 (100 μM; 30 min) significantly increases the spontaneous acrosome reaction rate in acrosome reaction assay performed on permeabilized mouse sperm[5].

trans-Ned 19 (20 mg/kg; i.p.; once administered 2 h before the first injection of anti-CD3 antibody and then once daily for 3 days or once administered 2 h before the injection of nephrotoxic serum and then once daily for 10 days) improves disease progression and reduces weight loss in a mouse model of transient intestinal inflammation induced by CD3-specific antibody, and improves renal damage in mice with nephrotoxic serum nephritis[4].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 6.25 mg/mL (12.15 mM); Clear solution
This protocol yields a clear solution of ≥ 6.25 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (62.5 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 6.25 mg/mL (12.15 mM); Clear solution
This protocol yields a clear solution of ≥ 6.25 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (62.5 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
