BRD3290
BRD3290 is a GluA3-preferring AMPA receptor positive allosteric modulator with brain exposure in Mus musculus (mice) following peripheral administration. BRD3290 preferentially potentiates GluA3-containing AMPA receptors over GluA1/2-containing AMPA receptors. BRD3290 increases cerebellar cGMP levels in Mus musculus (mice). BRD3290 does not induce convulsions, alter motor function, or improve novel object recognition task performance in Mus musculus (mice) at tested doses. BRD3290 can be used for the research of schizophrenia.

In FLIPR calcium flux assays, BRD3290 (0.068 μM; induced by 48 h Doxycycline (HY-N0565)) exhibits 10-fold higher activity in HEK293T cells expressing GluA3i/2o than in those expressing GluA1i/2o[1].
BRD3290 (10 nM-100 µM; 48 h Doxycycline induction, 3 min pre-incubation) enhances GluA3i/2o AMPAR with an EC50 of 3.7 µM; in automated patch-clamp electrophysiology assays using HEK293T cells, its activity is 10.9-fold higher than that of GluA1i/2o AMPAR (EC50 = 16.9 µM)[1].
