a15:0-i15:0 PE is an atypical TLR2 agonist, and its immune signaling strictly depends on the TLR2-TLR1 heterodimer. a15:0-i15:0 PE induces dendritic cells to produce TNFα and IL-6, but does not induce the production of IL-10 or IL-12p70. At low doses, a15:0-i15:0 PE resets the activation threshold of immune cells to suppress or regulate immune responses against specific agonists.

a15:0-i15:0 PE (50 μg/mL; overnight) induces TNFα and IL-6 release from murine bone-marrow-derived dendritic cells via a TLR2-dependent pathway, with no induction of IL-10 or IL-12p70[1].
a15:0-i15:0 PE (50 μg/mL; overnight) requires distinct branched-chain acyl groups for TNFα-inducing activity in murine bone-marrow-derived dendritic cells, with positional isomer i15:0-a15:0 showing partial activity[1].
a15:0-i15:0 PE (0.1-10 μg/mL; 3-21 h) induces TNFα release from human monocyte-derived dendritic cells via a TLR2-TLR1 heterodimer-dependent pathway, and low-dose pre-treatment for 18 h suppresses subsequent TNFα responses to Pam3CSK4 and moderates responses to LPS[1].
a15:0-i15:0 PE (50 μg/mL; 6 h) induces pro-inflammatory cytokine gene expression (TNFα, IL-6) in human peripheral blood monocytes, with limited induction of IL-23A and IL-12B[1].
