Ac-FR-AFC TFA (incubated for 1.5 h) serves as an effective fluorescent substrate for detecting cathepsin L activity in cell lysates of human glioblastoma U251 and U87 cells[1].
Cells are lysed after treatment with PBS (control group) or 5 μM TFP for 24 h. The supernatant is collected, mixed with the cathepsin L substrate (AC-FR-AFC), and incubated at 37°C for 1.5 h. Samples are then detected using a fluorescence microplate reader, with the excitation/emission wavelengths set at 400 nm/505 nm[1].
