Alicaforsen sodium
Alicaforsen (ISIS-2302) sodium is an oligonucleotide and immunostimulant targeting human ICAM-1 mRNA. Alicaforsen sodium hybridizes to specific sites to reduce the expression level of ICAM-1. Alicaforsen sodium is applicable to relevant research on psoriasis, rheumatoid arthritis and inflammatory bowel disease.

Animal Model:CD-I (6-8 weeks old, 20-30 g, male and female)[1]

Dosage:0.8 mg/kg; 4 mg/kg; 20 mg/kg; 100 mg/kg

Administration:i.v.; every other day; 27 days

Result:Increased incidence of injection-site hematomas at doses >4 mg/kg; observed injection-site hemorrhage, chronic inflammation, and perivasculitis at 20 and 100 mg/kg, with partial reversibility after a recovery period.
Triggered 3- to 4-fold increases in aspartate and alanine aminotransferase, 1.5- to 2-fold increases in total bilirubin and alkaline phosphatase, and reductions in cholesterol (41-48% of control), triglycerides (63-71% of control), and glucose (76-82% of control) at 100 mg/kg; these changes were mostly reversible after recovery, with only slight persistent elevations in alkaline phosphatase and aspartate aminotransferase.
Produced dose-dependent histopathologic changes.
