Antifungal agent-170
Antifungal agent-170 is an Antifungal agent. Antifungal agent-170 potently inhibits the growth of recombinant Saccharomyces cerevisiae strains expressing wild-type and mutant fungal CYP51 enzymes. Antifungal agent-170 potently inhibits the growth of Candida albicans, Candida glabrata, Candida krusei, Cryptococcus neoformans and Cryptococcus gattii. Antifungal agent-170 provides survival benefits in the Galleria mellonella (greater wax moth) infection model. Antifungal agent-170 can be used for the research of invasive fungal infections.

Antifungal agent-170 (Compound 15) potently inhibits growth of clinical isolates of Candida albicans, Candida glabrata, Candida krusei, Cryptococcus neoformans, and Cryptococcus gattii with MIC ranges of <0.004- >2 μg/mL, <0.004- >2 μg/mL, <0.004-2 μg/mL, 0.008-1 μg/mL, and 1-2 μg/mL, respectively, after 48 h incubation for Candida spp. and 72 h incubation for Cryptococcus species[1].
Antifungal agent-170 potently inhibits growth of recombinant Saccharomyces cerevisiae strains expressing wild-type and mutant fungal CYP51 enzymes, with MIC50 values ranging from <0.004 to 0.0313 μg/mL, and shows reduced activity against strains overexpressing ABC efflux pumps (MIC50 >2 μg/mL) compared to MFS efflux pumps (MIC50 0.125 μg/mL)[1].
Antifungal agent-170 exhibits rapid metabolic clearance in mouse, rat liver microsomes, with half-lives of 28.3 min, 8.76 min, respectively[1].
Antifungal agent-170 has low cytotoxicity against human A549 cells with an IC50 of 32 μg/mL after 24 h incubation, and a high selectivity index of 32,000 relative to its antifungal activity against Candida albicans SC5314[1].
