In Vitro

BI-1124 (0.075 mg/mL, 0.0075% w/v) exhibits high corneal permeability, sufficient solubility, and a suitable partition coefficient at pH 7.4 in in silico ADMET and ocular pharmacokinetic analyses. After simulated topical administration, its concentrations in rabbit cornea and conjunctiva can remain sustained above the in vitro IC50 (7 nM) and IC90 (70 nM) levels, which supports its potential to be repurposed for age-related dry eye disease through the inhibition of cathepsin S[1].
