Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs)[2].
1. Solution preparation
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 1-10 mM (optimized according to the experiment).
1.2 Preparation of working solution
Dilute to 1 nM-10 μM with buffer C (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Competition binding assays
2.1 HsHsp90 and PfHsp90 are added at final concentrations of 32 nM and 60 nM, respectively.
2.2 Assay mixtures contained 1 nM Geldanamycin-FITC in buffer C in 384-well microplates.
2.3 The final assay volume is 50 μL in all wells.
2.4 Plates are incubated overnight at 4 °C, followed by centrifugation.
2.5 The fluorescence polarization (FP) signal in each well is measured by use of a 480-nm excitation filter and a 535-nm emission filter on an EnVision system.
