BI-1910 is an agonistic human IgG2 monoclonal antibody targeting TNFR2, which selectively binds to human TNFR2 without inhibiting TNF-α binding. As an endogenous agonist, BI-1910 activates TNFR2 and mediates the activation of the NF-κB signaling pathway. Its agonistic activity is independent of FcγR or cross-linking; it can directly costimulate T cells and NK cells, induce T cell proliferation, and activate tumor-specific CD8+ T cells. BI-1910 exhibits antitumor activity in vivo, and shows an additive antitumor effect when combined with anti-PD-1 (HY-P9902). It can be used in research related to advanced/metastatic non-small cell lung cancer and hepatocellular carcinoma. Recommended isotype control: Human IgG2 lambda, Isotype Control (HY-P991206).

BI-1910 (15 min) acts as an agonist of NF-κB signaling in TNFR1-deficient NK-925 cells, with enhanced activity in the presence of TNF-α[1].
BI-1910 (10 μg/mL; 24 h) acts as an agonist to increase IFN-γ production from primary human NK cells[1].
BI-1910 (10 μg/mL; 72 h) acts as an intrinsic agonist to promote proliferation of activated primary human CD4+ T cells, with activity independent of cross-linking[1].
BI-1910 (10 μg/mL; 30 min pre-incubation, followed by 72 h co-culture) delivers TNFR2-mediated co-stimulatory activity to CD3/CD28-stimulated primary human T cells co-cultured with TAMs[1].
BI-1910 (6 h) activates NF-κB signaling in TNFR2-expressing Jurkat reporter cells, as measured by GFP expression[1].

Animal Model:C57BL/6J B-hTNFR2 (human TNFR2 knock-in) (female)
Female B-hTNFR2 knock-in mice (C57BL/6J background), 6-8 weeks old, were subcutaneously inoculated with 0.5 × 106 or 1 × 106 MC38 colon cancer cells suspended in 100 μL PBS into the flank; when tumors reached 40-200 mm3, the mice were randomized into treatment groups based on tumor size and treated via intraperitoneal injection at a dose of 40 mg/kg in a volume of 200 μL.
