Phosphofructokinase 1 (PFK1) is a glycolytic enzyme and also an allosteric activation target of fructose-2,6-bisphosphate. PFK1 supports cardiac glycolysis to maintain normal cardiac function and mediates adaptive responses to pressure overload. Upregulated expression and activity of PFK1 are associated with the alleviation of pathological changes induced by pressure overload, including diastolic dysfunction, interstitial fibrosis and myocardial hypertrophy. Inhibition of PFK1 redirects glucose carbon flux to the pentose phosphate pathway. PFK1 promotes proliferation, migration and glycolysis of colorectal cancer cells, reduces apoptosis and decreases their radiosensitivity, and shows higher expression levels in radioresistant tumors. PFK1 can be used in research related to diastolic dysfunction heart failure and colorectal cancer.

Phosphofructokinase 1 enables the quantitative detection of fructose-2,6-bisphosphate levels via a coupled-enzyme cell-free assay[1].
Phosphofructokinase 1 (PFK1) is expressed in a variety of human colorectal cancer (CRC) cell lines, with the highest expression level detected in HT29 cells; in addition, its expression level in radioresistant tumors is significantly higher than that in radiosensitive tumors[3].
Phosphofructokinase 1 knockdown via siRNA-PFK1 (24 h post-transfection) suppresses glycolytic activity in human colorectal HT29 and HCT116 cells, and lowers glucose consumption, lactate secretion, intracellular ATP content as well as PFK1 enzymatic activity[3].
Phosphofructokinase 1 knockdown via siRNA (transfection for 24 h; 24-72 h post-transfection) inhibits the proliferation and migration of human colorectal cancer cells HT29 and HCT116, and promotes their apoptosis[3].
Phosphofructokinase 1 knockdown via siRNA-PFK1 (transfection for 24 h; incubation for 24 h post-irradiation) enhances the radiosensitivity of human colorectal cancer HT29 and HCT116 cells, and reduces cell survival rates after X-ray irradiation at doses of 2 to 8 Gy, with an LD50 value of 6.2 Gy for HT29 cells and 4.2 Gy for HCT116 cells; it also enhances radiation-induced apoptosis of HT29 and HCT116 cells in a dose-dependent manner[3].
Phosphofructokinase 1 knockdown via siRNA-PFK1 (transfection for 24 h; incubation for 14 days after seeding) reduces the colony-forming ability of HT29 human colorectal cancer cells, and this effect is enhanced after exposure to 2 Gy X-rays[3].
