BacPROTAC-1 is a BacPROTAC-class degrader that recruits the ClpC:ClpP protease to induce target protein degradation. BacPROTAC-1 binds to monomeric streptavidin (mSA), Bacillus subtilis ClpCNTD and Mycobacterium smegmatis ClpC1NTD with Kd values of 3.9, 2.8 and 0.69 μM, respectively. BacPROTAC-1 forms a ternary complex between the substrate and ClpC, promotes ClpC reassembly and activation, and induces ATP-dependent degradation of mSA and mSA-fused model substrates via the ClpCP or ClpC1P1P2 protease system. BacPROTAC-1 can be used in studies of bacterial targeted protein degradation, bacterial protein homeostasis and antibacterial target discovery.

BacPROTAC-1 (400 μM; syringe load) binds purified Bacillus subtilis ClpCNTD with a KD of 2.8 μM[1].
BacPROTAC-1 (400 μM; syringe load) binds purified monomeric streptavidin (mSA) with a KD of 3.9 μM[1].
BacPROTAC-1 (400 μM; syringe load) binds purified Mycobacterium smegmatis ClpC1NTD with a Kd of 0.69 μM[1].

BacPROTAC-1 (1-100 μM; 2 h at 37°C) induces degradation of monomeric streptavidin (mSA) by reconstituted Bacillus subtilis ClpCP, with efficient degradation observed at 100 μM after 2 hours at 37°C[1].
BacPROTAC-1 (0.1-100 μM; 2 h at 37°C) potently induces degradation of mSA-Kre by reconstituted Bacillus subtilis ClpCP, with efficient degradation observed at 1 μM after 2 hours at 37°C, and this activity is ATP-dependent and blocked by biotin or pArg[1].
BacPROTAC-1 (1-100 μM; 2 h at 37°C) induces degradation of mSA-NrdI, mSA-TagD, and mSA-NusA by reconstituted Bacillus subtilis ClpCP, but with lower efficiency than mSA-Kre, after 2 hours at 37°C[1].
BacPROTAC-1 (156 μM) induces conversion of inactive decameric Bacillus subtilis ClpCDWB into an active tetramer of hexamers when bound to mSA-Kre[1].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (2.22 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (2.22 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
