LCK degrader-2
LCK degrader-2 is a selective CRBN-dependent molecular glue degrader targeting LCK. LCK degrader-2 induces the proximity effect between LCK and CRBN, stabilizes the CRBN-LCK ternary complex, and triggers the ubiquitination and degradation of LCK. LCK degrader-2 can be used for the research of acute lymphoblastic leukemia.

LCK degrader-2 (Compound 17) (5 µM) is a molecular glue that selectively stabilizes the CRBN-LCK ternary complex; in ASMS analysis using purified compounds, it shows a relative enrichment factor of 2.4-fold and does not stabilize the CRBN-GSPT1 ternary complex[1].
LCK degrader-2 induces the proximity between recombinant CRBN and LCK in AlphaScreen assays, with an EC50 in the double-digit nM range; at concentrations above 1×10-8 M, it reduces LCK protein levels to nearly undetectable levels[1].
