MP-RM-1 is an antibody-based, non-competitive ErbB-3 inhibitor with a Kd value of 32.7 nM. MP-RM-1 inhibits both ligand-dependent and ligand-independent ErbB-3 activation, blocks ErbB-2/ErbB-3 heterodimerization, induces ErbB-3 internalization and degradation, and suppresses the phosphorylation of downstream Akt. MP-RM-1 exerts its antagonistic effect through a non-competitive mechanism. MP-RM-1 inhibits tumor growth in melanoma and prostate cancer xenograft models in nude mice and reduces the proliferative activity of tumor cells.

Description

MP-RM-1 is an antibody-based, non-competitive ErbB-3 inhibitor with a Kd value of 32.7 nM. MP-RM-1 (0.01-10 μg/mL; 15-120 min) inhibits NRG-1β-dependent phosphorylation of ErbB-3 and Akt in a dose- and time-dependent manner in breast cancer cells MDA-MB-435 and melanoma cells IR-8[1].
MP-RM-1 (10 μg/mL) significantly inhibits the colony-forming ability of IR-8 melanoma cells[1].

In Vivo

MP-RM-1 (20 mg/kg; i.p.; twice weekly) significantly inhibits the growth of ErbB-3 melanoma xenografts in nude mice, and reduces ErbB-3 expression and Akt phosphorylation levels in tumor tissues[1].
MP-RM-1 (20 mg/kg; i.p.; twice weekly) significantly inhibits the growth of DU-145 prostate cancer xenografts in nude mice[1].
