Penehyclidine (Penequinine) hydrochloride, a anticholinergic agent, is a selective antagonist of M1 and M3 receptors. Penehyclidine hydrochloride activates NF-kβ in lung tissue and inhibits the release of inflammatory factors. Penehyclidine hydrochloride can alleviate the pulmonary inflammatory response in rats with chronic obstructive pulmonary disease (COPD) undergoing mechanical ventilation.

In Vitro
Penehyclidine (0.01-1.0 μM, 2 h) reduces the levels of IFN-γ, TNF-α, IL-1β, IL-6, promotes LPS (HY-D1056)-induced cell proliferation of BEAS-2B, inhibits the apoptosis, and promotes the LPS (HY-D1056)-induced autophagy in BEAS-2B[2].
Penehyclidine (0.1 μM, 2 h) activates the Akt/GSK-3β signaling pathway, and alleviates the A/R-induced cell injury in H9c2 cells[3].

In Vivo
Penehyclidine (1 mg/kg, ip, single dose) enhances the expression of BDNF and phosphorylation of ERK1/2 in the hippocampus, exhibits antidepressant-like effects in mouse CUMS model[4].
