QX008N (2 μg/mL initial concentration, 1:5 gradient dilutions) specifically binds human, cynomolgus monkey, and rhesus macaque TSLP[1].
QX008N potently inhibits the interaction between human TSLP and the TSLPR-IL-7Rα complex[1].
QX008N (2 μg/mL initial concentration, 1:3 gradient dilutions) potently neutralizes TSLP-induced JAK/STAT signaling in SW756-STAT5-Luciferase cells exposed to recombinant human, natural human, or recombinant cynomolgus monkey TSLP[1].
QX008N (10 concentration points via 1:5 gradient dilutions) potently inhibits human TSLP-induced CCL17 secretion in human PBMCs (IC50 = 9.836 ng/mL) and whole blood (IC50 = 1.599 ng/mL), and CCL22 secretion in human PBMCs, with activity superior to the Tezepelumab analog[1].
QX008N, when combined with anti-IL-4R antibody Oturkibart (HY-P991152), potently inhibits CCL17 production in human PBMCs stimulated with TSLP, IL-4, IL-13, and IL-33, achieving 95.5% inhibition that exceeds the additive effect of monotherapies[1].
QX008N (6 day incubation), when combined with anti-IL-4R antibody QX005N, enhances inhibition of OVA-induced CCL17, IL-5, and CCL26 production in a ternary culture model of MRC-5 fibroblasts, A549 epithelial cells, and human PBMCs, compared to QX008N monotherapy[1].

In Vivo

QX008N (10-50 mg/kg; once weekly; 6 doses; subcutaneous injection) dose-dependently improves lung function, reduces peripheral blood and bronchoalveolar lavage fluid inflammatory markers, and decreases eosinophil infiltration in a cynomolgus monkey asthma model[1].

Animal Model: Cynomolgus monkey (male, 3-6 years, 2.5-7.0 kg, sensitized via intraperitoneal injection of Ascaris suis extract mixed with Al(OH)3 adjuvant at weeks 1, 2, and 3, followed by nebulized allergen challenge at weeks 4-7)[1]
