VtCD-cTP-ALKF-2
VtCD-cTP-ALKF-2 is a DAPK1 PROTAC degrader. VtCD-cTP-ALKF-2 can bind to dephosphorylated DAPK1, recruit VHL E3 ligase, and induce ubiquitin-proteasome-mediated degradation. VtCD-cTP-ALKF-2 can be used for research on Alzheimer's disease.
(Pink: DAPK1 ligand (HY-P12376); Blue: VHL ligand (HY-125845); Black: linker (HY-22408)).

Biological Activity
Description
In Vitro
VtCD-cTP-ALKF-2 (10 μM; 12-48 h washout monitoring), generated from its caged precursor, induces ROS- and proteasome-dependent DAPK1 degradation with 62% reduction of total DAPK1 and downstream reduction of p-APP and p-Tau in Aβ42 oligomer-induced SH-SY5Y cells[1].
VtCD-cTP-ALKF-2 (100 μM H2O2; 24 h) is generated from its caged precursor with near-complete conversion and remains stable at pH 7.4 without H2O2[1].

Alzheimer’s Disease Modeling
Alzheimer’s Disease (AD) is a neurodegenerative disorder characterized by a progressive decline in cognitive functions and loss of specific types of neurons and synapses. Alzheimer's symptoms can be simulated in mice by injecting drugs (such as Aβ) or genetically modified.
