Zigakibart (BION-1301) (0.3-30 mg/kg; i.v.; single dose) produces a dose-dependent reduction in circulating IgA and IgM with minimal impact on IgG in healthy female cynomolgus monkeys after a single intravenous dose[1].

Dosage: 0.3 mg/kg; 3 mg/kg; 30 mg/kg
Administration: i.v.; single dose
Result: Reduced circulating IgA and IgM in a dose-dependent fashion, with the 30 mg/kg dose producing the most substantial reductions.
Showed minimal change in circulating IgG levels across all doses.
Achieved statistically significant reductions in IgA and IgM relative to controls at 4 weeks after dosing.

Biological Activity
  • Immobilized APRIL/TNFSF13 Trimer Protein, Human (HEK293, Flag-His, HY-P70733) can bind Zigakibart. The EC50 for this effect is 7.098 ng/mL.
  • Flow cytometric analysis of 1.5X106 RAW264.7 cells treated with 1µg/mL LPS for 24h with Zigakibart (HY-P990070, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃, Human IgG4 kappa Isotype Control (HY-P99003, blue) was used as the isotype control，cells without incubation with primary antibody were used as the unlabeled control (black).
