    Description

    HLCL-61 hydrochloride is a first-in-class inhibitor of protein arginine methyltransferase 5 (PRMT5).

    IC50 & Target

    Protein arginine methyltransferase 5[1]

    In Vitro

    HLCL-61 hydrochloride (1-100 μM; 24-72 hours) reduces cells growth at dose-dependent manner with IC50s of 14.12, 16.74, 6.3, 8.72 μM for MV4-11 cells, THP-1 cells, FLT3-WT blast, and FLT3-ITD blast, respectively[1].
HLCL-61 hydrochloride shows effective inhibition of symmetric arginine dimethylation (me2) of histones H3 and H4 in AML samples, starting at 12 h post-treatment and persisting after 48 h[1].

MCE has not independently confirmed the accuracy of these methods. They are for reference only.

    Cell Viability Assay[1]

Cell Line: MV4-11 cells, THP-1 cells, FLT3-WT blast (primary blasts from patients), FLT3-ITD blast (primary blasts from patients)
    Concentration: 1, 5, 10, 25, 50, 100 μM
    Incubation Time: 24, 48, 72 hours
Result: Dose-dependent reduction in cell viability with IC50s of 14.12, 16.74, 6.3, 8.72 μM for MV4-11 cells, THP-1 cells, FLT3-WT blast, and FLT3-ITD blast, respectively.

    Help & FAQs
    • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
