Prostaglandin Receptor Control

Latanoprostene bunod (NCX116; LBN) is a nitric oxide-releasing prostaglandin F₂α analog. Latanoprostene bunod is a prodrug that, upon instillation into the eye, is hydrolyzed by corneal esterases into two active metabolites: Latanoprost (HY-B0577) and NO. Latanoprost activates the prostaglandin FP receptor to increase the outflow of aqueous humor through the uveoscleral pathway. NO increases aqueous humor drainage through the trabecular meshwork pathway, achieving synergistic enhancement targeting the dual pathways of aqueous humor outflow. Latanoprostene bunod can be used in research related to open-angle glaucoma and ocular hypertension.

Tafluprost (AFP-168) is an anti-glaucoma prostaglandin (PG) analog. Tafluprost can inhibit the apoptosis of retinal ganglion cells (RGCs) and rat RGCs cells. Tafluprost promotes axon regeneration by regulating Zn²⁺-mTORpathway, inhibits intracellular lipid accumulation in human preorbital adipocytes. Tafluprost can be used in the study of optic nerve injury in glaucoma.

Thromboxane B2 is a prostaglandin derivative that is released during anaphylaxis. Thromboxane B2 induces arterial contraction and platelet aggregation. Thromboxane B2 is a biological inactive product and circulating catabolite of thromboxane A2. Thromboxane B2 levels in peripheral venous blood and coronary sinus blood can be used as a diagnostic marker for angina. Thromboxane B2 level in serum is also associated with inhibition of COX-1 activity inhibition in platelets. Thromboxane B2 is a major product of prostaglandin endoperoxide metabolism. Thromboxane B2 produces dose-related decreases in airflow rate, tidal volume and dynamic lung compliance, whilst simultaneously increasing pulmonary airway resistance in dog model. Thromboxane B2 appears to be a naturally occurring bronchoactive metabolite in the bioconversion of arachidonic acid.

Crisdesalazine (AAD-2004) is a microsomal prostaglandin E₂ synthase-1 (mPGES-1) inhibitor. Crisdesalazine acts as a potent free radical scavenger that directly neutralizes reactive oxygen species (ROS) including hydrogen peroxide, exerting neuroprotective effects against apoptosis and axonal damage. Crisdesalazine inhibits PGE₂ production, mediates inflammatory responses, and promotes the conversion of macrophages from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype. Crisdesalazine is applicable to neuroprotection research in multiple sclerosis and spinal cord injury.

U-44069 is a stable prostaglandin (PG) H2 analogue and a potent vasoconstrictor. U-44069 induces Ca influx at preglomerular vessels.

trans-Latanoprost (5,6-trans-Latanoprost) is the trans isomer of Latanoprost (HY-B0577). trans-Latanoprost is promising for research of glaucoma and high eye pressure.

1a,1b-Dihomo prostaglandin E1 (1a,1b-Dihomo PGE1) is an E1-type prostaglandin. 1a,1b-Dihomo prostaglandin E1 can be used for research on platelet aggregation.

15-deoxy-Δ12,14-Prostaglandin J2 glutathione (15-deoxy-Δ12,14-PGJ2 glutathione) is a non-enzymatic adduct formed from 15-deoxy-Δ12,14-PGJ2 and glutathione. The biological properties of this compound have not been characterized.

(±)5-iPF2α-VI (5-iso Prostaglandin F2α-VI) is the racemate of 5-iPF2α-VI. 5-iPF2α-VI is a regioisomeric isoprostane formed from arachidonic acid (AA) and is a biomarker of oxidative stress.

9-keto latanoprost is a derivative of the F-prostaglandin (FP) receptor agonist Latanoprost (HY-B0577).
