Anti-Human/Mouse CD95 Antibody (HFE7A) modulates the Fas-mediated apoptotic signaling pathway without blocking the binding of Jo2 to Fas. Anti-Human/Mouse CD95 Antibody (HFE7A) inhibits Jo2-induced caspase activation, mitochondrial depolarization, hepatocyte death and apoptosis. Anti-Human/Mouse CD95 Antibody (HFE7A) protects BALB/c mice against Jo2-induced acute liver injury and reduces Jo2-associated elevation of serum transaminase levels. Anti-Human/Mouse CD95 Antibody (HFE7A) (0.05-5 μg/mL; 1.5-6 h) inhibits Jo2-induced death of primary BALB/c mouse hepatocytes in vitro in a dose-dependent manner, as well as the activation of caspase-3, -8 and -9[1].

In Vivo

Anti-Human/Mouse CD95 Antibody (HFE7A) (16.5-200 μg; i.v.; single dose) completely rescues BALB/c mice from Jo2-induced lethal acute fulminant hepatitis and inhibits Jo2-induced hepatocyte apoptosis via suppression of caspase activation and mitochondrial depolarization[1].

Dosage: 67 μg (survival; AST/ALT inhibition); 16.5 μg (caspase suppression); 200 μg (mitochondrial depolarization reduction)
Administration: i.v.; single dose
Result: Achieved 100% survival (3/3 mice) at 4 and 24 hours post-Jo2 injection.
Inhibited Jo2-induced increases in serum AST and ALT levels.
Almost completely suppressed Jo2-induced activation of caspases-3, -8, and -9 in hepatocytes to basal levels.
Reduced the percentage of hepatocytes with mitochondrial depolarization when administered simultaneously with Jo2 at 200 μg.
