Arsthinenol  (Synonyms: Arsthinol; Balarsen)

Arsthinenol (Arsthinol; Balarsen) is an orally active small organic arsenic molecule. Arsthinenol exhibits anti-amoebic activity by eliminating cysts and trophozoites of Entamoeba histolytica in feces and promoting the healing of amoebic rectal ulcers. Arsthinenol forms the hydroxypropyl-β-cyclodextrin inclusion complex STB-HPβCD, which has an IC₅₀ of 31.6 μM against U87 MG cells and possesses anti-tumor activity. Arsthinenol can be used in the research of intestinal amoebiasis and glioblastoma^[1][2].

Arsthinenol (STB) forms 1:1 inclusion complexes with HPβCD and RAMEβCD, with high apparent stability constants measured via phase solubility studies at 25 °C^[2].
Arsthinenol (0.01 μM, 1 mM; 24-48 h) and its HPβCD complex exhibit dose- and time-dependent cytotoxicity against U87 MG glioma cells, with Arsthinol showing a significantly higher therapeutic index than reference arsenical compounds after 48 h incubation, reaching an IC₅₀ of 2.8 μM^[2].

Arsthinenol (STB) (0.12-1.2 mmol/kg; i.p., p.o.; 5 days/week) complexed with cyclodextrin via intraperitoneal administration at 0.12 mmol/kg 5 days/week significantly improves median survival to 21 days in a heterotopic mouse glioma model, with no observed toxicity, while oral administration at 1.2 mmol/kg 5 days/week does not yield statistically significant survival benefits relative to controls^[2].
