SMA-245

SMA-245 is an inhibitor of Filovirus glycoproteins, acting on Ebola virus (EBOV) and Marburg virus (MARV). SMA-245 blocks the endosomal membrane fusion step of filoviruses by binding to the EBOV GP1/GP2 fusion loop. SMA-245 inhibits MARV viral entry. SMA-245 can be used in research related to filovirus infections.

SMA-245 (compound 41) (48 h) potently inhibits pEBOV entry in A549 cells with an EC₅₀ of 0.040 μM, also inhibits pMARV, pSUDV, and pTAFV entry, shows no activity against pBDBV, and has low activity against non-filovirus pseudoviruses^[1].
SMA-245 (40 h infection incubation; 48 h cytotoxicity incubation) potently inhibits infectious EBOV replication in HeLa cells with an EC₅₀ of 0.04 μM and infectious MARV replication with an EC₅₀ of 0.38 μM, with selectivity indices of 120 and 12, respectively^[1].
SMA-245 moderately inhibits hERG potassium channels with an IC₅₀ of 2.93 μM, representing a potential cardiac safety concern^[1].
SMA-245 has excellent plasma stability in human and mouse, high stability in human liver microsomes, moderate stability in mouse liver microsomes, and low to moderate inhibition of major CYP450 isoforms at 10 μM^[1].
