2-5A
2-5A is a RNase L activator, signal mediator and innate immune second messenger. 2-5A induces the expression of interferons and interferon-stimulated genes in recipient cells in a RNase L-dependent manner. 2-5A inhibits tumor growth in mouse models via paracrine RNase L activation derived from cancer cells. 2-5A can be used in studies related to viral infection and cancer.

2-5A induces paracrine RNase L activation in co-cultured A549 human lung adenocarcinoma epithelial cell lines, with activation triggered by poly(I:C) treatment or expression of the constitutively active OAS1A76V mutant[1].
2-5A is exported extracellularly by poly(I:C)-stimulated MC38 murine colon adenocarcinoma cells via the ABCC10 and ABCC1 transporters[1].
Trimeric 2-5A is taken up by HEK293T human embryonic kidney cells and BJ1 human foreskin fibroblasts to induce RNase L-dependent IFN/ISG activation, with LRRC8A and P2X7R contributing to 2-5A import in HEK293T cells[1].
2-5A export via gap junctions (A549 cells) or ABCC10 (MC38 cells) mediates paracrine OAS-RNase L activation and antiviral immunity against VSV and SeV infection[1].
