TFLLRN-NH2 is a PAR-1 peptide-selective agonist. TFLLRN-NH2 induces AMPK phosphorylation and attenuates LPS-induced osteoclastogenesis, NFATc1 expression, NF-κB activation, and IκBα degradation. TFLLRN-NH2 stimulates MAPK phosphorylation in hepatocellular carcinoma cells. TFLLRN-NH2 induces concentration-dependent contraction of isolated human renal artery rings, stimulates IL-6 release from human T cells, and also evokes intracellular Ca2+ mobilization in primary human renal tubular epithelial cells. TFLLRN-NH2 can be used in research related to hepatocellular carcinoma and acute lung injury.

TFLLRN-NH2 (100 μM; 20 min) activates p42/p44 MAPKinase phosphorylation in HEP-3B cells[2].
TFLLRN-NH2 (100 μM) induces EGCG-sensitive p42/p44 MAPKinase activation in PHC1 and PHC2 primary HCC cultures[2].
TFLLRN-NH2 induces a significant, concentration-dependent contractile effect in isolated human renal artery rings[4].
TFLLRN-NH2 activates PAR-1 and induces intracellular Ca2+ mobilization in primary human tubular epithelial cells (HTECs); this peptide does not cross-activate PAR-2, cannot mediate PAR-1 transactivation of PAR-2, and fails to induce tissue factor (TF) synthesis in HTECs[5].
