Dephospho-CoA is a key intermediate in the biosynthesis of coenzyme A (CoA), which is catalyzed to form CoA by GTP-dependent Dephospho-CoA kinase (DPCK). Dephospho-CoA completes the final synthetic step of CoA via phosphorylation and participates in energy metabolism and cellular signal transduction. Dephospho-CoA can be used in studies involving cancer (e.g., regulation of cell proliferation) and metabolic diseases (e.g., mitochondrial dysfunction).

Dephospho-CoA (10 min) is quantitatively phosphorylated to CoA by DPCK, as confirmed by reverse-phase HPLC analysis of reaction products[1].
Dephospho-CoA (1 mM; 80 °C incubation for standard/kinetic assays; 70 °C incubation for specificity assays; 2 h preincubation for thermostability assays) was specifically phosphorylated to CoA by purified recombinant Thermococcus kodakarensis TK1697 protein, a GTP-dependent DPCK with optimal activity at 80 °C and pH 8.0, high thermostability, and a catalytic efficiency (kcat/Km) of 40.4 s−1 mM−1 for dephospho-CoA[2].
