Mitraphylline

Mitraphylline is the major pentacyclic oxindolic alkaloid presented in Uncaria tomentosa. Mitraphylline inhibits lipopolysaccharide-mediated activation of primary human neutrophils. Mitraphylline exhibits antiproliferative activity in human glioma and neuroblastoma cell lines. Mitraphylline can inhibit interleukin release in vivo.

In Vitro
Mitraphylline (25 μM) significantly reduces the NO production in human LPS-activated neutrophils[1].
Mitraphylline (25 μM, 6 h) significantly downregulates the pro-inflammatory cytokines in LPS-treated neutrophils[1].
Mitraphylline (0-40 μM, 48 h) inhibits cell viability dose-dependently in both GAMG (IC50 = 20.05 μM) and SKN-BE cells[3].

Cell Viability Determination by MTT Colorimetric Assay
The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
