Frexalimab (SAR441344) is a second-generation monoclonal antibody targeting the CD40 ligand (CD40L) with a good safety profile. Frexalimab inhibits the binding between CD40 and CD40L to modulate immune response. Frexalimab is likely to help prevent the process of β-cell destruction. Frexalimab is proming for multiple sclerosis, lupus erythematosus, Sj gren’s syndrome and type I diabetes research.

Frexalimab is a second-generation anti-CD40L humanized IgG1 monoclonal antibody that has been Fc-engineered. The Fc engineering abrogates the platelet activation that is triggered by FcγRIIa stimulation by immune complexes[2].

Flow cytometric analysis of 1X10EC6 500 ng/mL anti-CD3 stimulated Jurkat cells(24h) with Frexalimab (HY-P99625, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -
Immobilized Human CD40 Ligand/TNFSF5 Protein (His Tag) can bind Frexalimab. The EC50 for this effect is 2.43 ng/mL. -
Loaded Frexalimab on AHC2 biosensor, can bind CD40L/CD154/TRAP Protein, Human (HEK293, Tag free, HY-P70609) with an affinity constant of 4.117E-10 M as determined in BLI assay.
