Biological Activity
FWG-33B (5 μM; 4 hours) engages ~40% of total TRMT112 and >90% of METTL5-complexed TRMT112 in HCT116TRMT112-FLAG:HA-METTL5 cells[1].
FWG-33B (0.01-50 μM; 3 hours pretreatment) stereoselectively engages TRMT112 in HCT116TRMT112-FLAG:HA-METTL5 cells with an IC50 of 1.0 μM[1].
FWG-33B (40 μM; 30 minutes) enhances the methyltransferase activity of WT-TRMT112:METTL5 (but not TRMT112C100A:METTL5) by ~2-fold with 12-nt and 60-nt rRNA substrates[1].
FWG-33B (40 μM; 30 minutes) increases the catalytic efficiency of WT-TRMT112:METTL5 by reducing the Km for rRNA substrate from 10 μM to 4.7 μM[1].
