Cycotiamine is an S-acylated thiamine derivative with antiplasmodial activity. Cycotiamine inhibits the proliferation of Plasmodium falciparum and Plasmodium knowlesi. Cycotiamine can be used in malaria-related research.

Biological Activity
Cycotiamine (compound 12) exhibits IC50 values of 123 μM, 127 μM, and 176 μM against P. falciparum under extracellular thiamine concentrations of 0 μM, 2.97 μM, and 297 μM, respectively. The IC50 of cycotiamine against P. knowlesi exceeds 1000 μM, showing weaker anti-parasitic activity than other thiamine analogs in the same series. The IC50 of cycotiamine against human fibroblast HFF cells exceeds 1000 μM, with no obvious cytotoxicity[1].
Cycotiamine (serial dilution; 96 h) inhibits the proliferation of both Plasmodium falciparum strain 3D7-GFP and PfTPK-GFP-overexpressing Plasmodium to the same extent, indicating that its anti-Plasmodium effect is independent of thiamine pyrophosphokinase[1].
