Metamizole sodium (Dipyrone) is an orally active cyclooxygenase (COX) inhibitor. Metamizole sodium can inhibit cell proliferation and promote cell apoptosis. Metamizole sodium (1 and 10 μM, 24 h) can inhibit osteoblast differentiation and thus reduce its ability to generate new bone mineralized extracellular matrix[2].
Metamizole sodium (1-500 μM, 48 h) can inhibit cell proliferation and promote apoptosis in pancreatic cancer cell lines PaTu 8988 t and Panc-1[3].

Result: Inhibited proliferation in the PaTu 8988 t cell line with 1–500 μM and with concentrations of 1 μM, 10 μM, 100 μM, and 250 μM in the Panc-1 cell line.

In Vivo

Metamizole sodium (500 mg/kg, intraperitoneal injection twice a day for 7 days) can reduce the development of neuropathic pain symptoms in rats[4].
Metamizole sodium (250 or 500 mg/kg, orally) can effectively relieve pain and oxidative stress induced by scalpel incision and carrageenan in rats[5].

Animal Model: Male Wistar rats (270-300 g), with peripheral neuropathic pain[4]
Dosage: 500 mg/kg
Administration: Intraperitoneal injection, twice a day, for 7 days
Result: Diminished the expression of pronociceptive interleukins (IL-1beta, IL-6, and IL-18) and chemokines (CCL2, CCL4, and CCL7) in DRG measured 7 days after sciatic nerve injury.
Animal Model: Pain model in rats [5]
Dosage: 250 or 500 mg/kg
Administration: p.o.
Result: Decreased the COX-2 gene expression at 500 mg/kg and increased the MPO gene expression. Reduced MDA levels in the carrageenan paw test and increased tGSH levels.
